ABSTRACT

Objective:To explore the changing characteristics of interpretive bias of senior high school students with different levels of autistic traits in ambiguous situations, and to investigate the consistency of interpretive bias across self/other relevant conditions.Methods:A survey of 1 080 senior high school students from a high school in Anhui province was conducted by using the autistic-spectrum quotient (AQ). According to the criteria of high scores and low scores of 27%, the subjects in the high autistic trait group ( n=266) and the low autistic trait group ( n=266) were selected.The ambiguous situation paradigm was used to measure the frequency of positive interpretation of ambiguous information by two groups of subjects under the conditions of self and others, SPSS 26.0 was used for descriptive statistics, and jamovi 2.2.3 was used for generalized linear mixed model analysis. Results:(1) The results of the between-group effects at the level of autistic traits indicated that the frequency of positive interpretations in the high autistic group was significantly lower than that in the low autistic group(54.5(11.8), 57(11.8) )( χ2=13.99, P<0.001). The results of the interaction between level of autistic traits and type of ambiguous situation indicated that in the positive ambiguous situation, the frequency of positive interpretations in the high autistic group was smaller than that in the low autistic group (33(4), 34(3)) ( P<0.001). There was no significant differences in the number of positive interpretations between high autistic group and low autistic group in negtive ambiguous situation( P>0.05). (2) The results of the main effects of the ambiguous situation types indicated that the frequency of positive interpretations in positive ambiguous situations was significantly higher than that in negative ambiguous situations(33(4), 23(9.25)) ( χ2=1 348.50, P<0.001). The results of the interaction between level of autistic traits and type of ambiguous situation indicated that the frequency of positive interpretations in positive ambiguous situations (33(4), 34(3)) in both high and low autistic groups was larger than that in negative ambiguous situations (22(10), 24(9)) ( P<0.001). (3) The main effects results for the self/other related conditions suggested that the frequency of positive interpretations of familiarity with others (19(4)) was significantly higher than that of self-related conditions (19(5)) and strangers (19(5)) ( χ2=9.51, P<0.01), and there was no significant difference between self-related conditions and strangers( P>0.05). The results of the interaction between ambiguous situation type and self/other related conditions suggested that in the positive ambiguous situation, the frequency of positive interpretations of familiarity with other people's conditions was greater than that of self-related conditions( P<0.01), and in the negative ambiguous situations, there was no significant difference in the frequency of positive interpretations related to different self/others( P>0.05). Conclusions:(1) High school students with high and low autistic traits are more inclined to show positive interpretations in the ambiguous situations, and it higher under the condition of positive ambiguous situations and acquaintances.(2) Compared with those with low autistic traits, individuals with high autistic traits tend to give less positive interpretations to ambiguous situations, but this difference is mainly manifested in positive ambiguous situations.(3) In the negative ambiguous situation, there is no significant difference in number of positive interpretations produced by senior high school students with high and low autistic traits, and they are stable across self and other related conditions.

ABSTRACT

AIM:To investigate the neuroprotective effect of 17β-estradiol(E2)on retina light damage in BALB/c and C57BL/6 mice and provide experimental data for the successful construction of a research model for E2 against retinal light damage.METHODS:Totally 40~45 adult female BALB/c or C57BL/6 mice were divided into six groups, 6 for each group: normal control, ovariectomized control, ovariectomized light(mice were stimulated with continuous white light at 10000 lx for 4, 8, 12, 16, and 24h after 14d of ovariectomy), intravitreal administration sham operation, saline and E2 pre-treatment groups(2μL saline or 10-5mol/L E2 were intravitreal injected respectively after 14d of ovariectomy operation and 24h of dark adaptation). The morphological and functional changes of the retina were detected by paraffin section HE staining, TUNEL staining and electroretinogram.RESULTS:In the ovariectomized light group, the thickness of the inner/outer nuclear layer decreased significantly from the 4h stimulation of 10000 lx white light group. Intravitreal administration of E2 significantly inhibited the apoptosis of retinal cells in the two strains of mice(P&#x003C;0.01)and the decrease of amplitudes of a- and b-waves in max-ERG of C57BL/6 mice(P&#x003C;0.05).CONCLUSION:The light loss sensitivity of two strains of mice was different under the same light stimulation. E2 had a protective effect on both morphology and function of the retina in BALB/c mice, and had a significant protective effect on retina function in C57BL/6 mice.

ABSTRACT

Objective To detect the pre- and post-treatment levels of serum interleukin (IL-20) in asthmatic patients during the acute exacerbation period and to investigate the role of IL-20 in the pathogenesis of bronchial asthma and relative clinical significance. Methods Forty-five cases of mild to moderate asthma outpatients in the First Affiliated Hospital of Guangxi Medical University from May 2013 to October 2013 were chosen as the asthma group and 32 healthy people who underwent routine physical examination as the control group. All the patients were treated with inhalation salmeterol and fluticasone 50/250 μg/μg (sucking twice a day) together with ventolin if necessary , and even with theophylline sustained-release tablets as the additional treatment in moderate asthma group for 1 week. The serum levels of IL-20 as well as immunoglobulin E (IgE) were detected by ELISA in the asthma group before and after treatment as well as in the control group. Other data including ECP , the number of eosinophil ceils (EOS) and the pulmonary function (FEV1%) were detected as well. The differences of IL-20 levels between the asthma group before and after treatment and the control group were analyzed and the correlation between IL-20 and Ig E, EOS, ECP and FEV1% were analyzed. Results Compared with the healthy control group [(13.58 ± 6.17)pg/L], the levels of IL-20 in the mild and moderate asthma pretreatment group were significantly increased [(23.43 ± 13.60)pg/L and (33.78 ± 22.69)pg/L]. Compared with the mild asthma group, the levels of IL-20 in the moderate asthma pretreatment group were significantly higher. After treatment , the levels of IL-20 [(15.73 ± 8.27)pg/L and (19.64 ± 11.69)pg/L] were decreased respectively in the asthmatic patients. The Pearson correlative analysis showed that there were positive correlations between the levels of 1L-20 and IgE , EOS and ECP respectively and there were negative correlations between IL-20 levels and FEV1%. Conclusion IL-20 may be involved in the pathogenesis of asthma.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the antifibrotic effect of oxymatrine and captopril in mice with chronic viral myocarditis (CVMC) and determine the possible antifibrotic mechanism of oxymatrine in CVMC.</p><p><b>METHODS</b>Ninety Balb/c mice were randomly divided into normal control group 1 (n = 10), normal control group 2(n = 10) and CVMC model group (n = 70). The mice in CVMC model group were infected with coxsackievirus B(3) (CVB(3)) on days 0, 14 and 28 to establish CVMC model. The volume of CVB(3) suspension was 0.20 ml, 0.25 ml and 0.30 ml, whose 50% tissue culture infection dose was 10(9) respectively. The mice in the normal control group 1 and 2 were given normal saline of volumes equal to those of viral suspension given to the model group at the same time points. Echocardiography and collagen specific picrosirius red staining were performed to evaluate the CVMC model on day 42 for the mice of the normal control group 1 and 8 mice of CVMC model group. The remaining mice in CVMC model group were randomly divided into CVMC control group, captopril group and oxymatrine group on day 42. From then on, the mice in captopril group and oxymatrine group were treated with captopril or oxymatrine at the dose of 100 mg/kg, by gavage once a day for 28 days, and meanwhile the mice in CVMC control group and the normal control group were given equal-volume normal saline by gastric gavage every day, for 28 days successively. All these mice were sacrificed on day 70. Heart tissue slices were stained with collagen specific picrosirius red and the collagen volume fraction (CVF) was calculated with image analysis software. The expressions of AngII and TGF-beta1 were determined by immunohistochemistry and Western blotting.</p><p><b>RESULTS</b>Compared with normal group 1, the left ventricular end-diastolic internal diameters, left ventricular end-systolic internal diameters and heart rates were significantly increased in CVMC model group (P < 0.05, P < 0.01, P < 0.05, respectively), ejection fractions, fractional shortenings and peak velocity of aorta were all significantly decreased in CVMC model group (P < 0.01 for all comparisons), and CVF levels were significantly increased in CVMC group (P < 0.01) on day 42. Compared with normal control group 2, captopril group and oxymatrine group, CVF levels and the expressions of TGF-beta1 were significantly increased in CVMC control group (P < 0.01 for all comparisons) on day 70. The expressions of AngII in CVMC control group were higher than those in normal control group and captopril group (P < 0.01 for all comparisons), but there were no significant difference between oxymatrine group and CVMC control group (P > 0.05) on day 70.</p><p><b>CONCLUSION</b>Oxymatrine can inhibit myocardial fibrosis in CVMC, and the mechanisms of its antifibrotic effects might be related with the down-regulation of TGF-beta1 expression.</p>

ABSTRACT

To investigate the effects of microenvironment of aorta-gonad-mesonephros (AGM) on embryonic hematopoiesis, mesenchymal stem cell like stromal cells (MSC like stromal cells) derived from dorsal aorta (DA) in AGM region were separated and identified by their growth characteristics, related molecules expression and mesenchymal lineage potentials. Stromal cells from DA region in mouse embryos (E11.5) were isolated and cultured in vitro. After transfected by pSV3neo-SV40, the clones with G418 resistance were selected, and their growth characteristics were studied. The related molecules were analyzed by flow cytometry, and each clone was induced to differentiate into adipocytes, osteocytes, and chondrocytes. The results showed that most clones (20 clones) selected in the mouse DA region held the morphology of fibroblastoid cells. mDAF3 and mDAF18 could be grown in culture for more than 50 passages with G418 resistance, both have the potential to differentiate into adipocytes, osteocytes, and chondrocytes. At the logarithmic growth period, the cell population doubling time is about 24 hours. Surface markers, such as CD29, CD44, CD105 and Sca-1 were positively detected, while low levels of CD34, CD45, and CD31 were detected. It is concluded that immortalized mDAF3 and mDAF18 have the specific phenotype and differential potency of MSC, which suggests that MSC maybe exist in mouse embryonic DA region, where the MSC like stromal cells can be used as a cell model for further research on the modulation activity of DA microenvironment for embryonic hematopoiesis.

ABSTRACT

Objective To observe the protective function of murine compact bone derived-mesenchymal stem cells(MSCs) on bleomycin-induced lung injury.Methods MSCs were isolated from the mouse compact bone and then cultured.The immunophenotype and differentiation potential of MSCs were identified by flow cytometry and inductive culture.56 female C57BL/6 mice were assigned into eight groups(7 each): normal group,busulfan intraperitoneal administration group(BUS),bleomycin-induced lung injury group(BLM),bleomycin-induced lung injury and busulfan intraperitoneal administration group(BLM+BUS),BLM challenge and MSCs treatment group(BLM+MSCs),BLM challenge and busulfan and MSCs administration group(BLM+BUS+MSCs).According to infused cell number,BLM+MSCs and BLM+BUS+MSCs group were again respectively divided to two subgroups: BLM+MSCs 1 group(5?105 MSCs) and BLM+MSCs 2 group(2.5?105 MSCs),and BLM+ BUS+MSCs 1 group(5?105 MSCs) and BLM+BUS+MSCs 2 group(2.5?105 MSCs),respectively.The mice were sacrificed 14 days after MSCs infusion,and their lungs were removed for pathological analysis and quantification of hydroxyproline.Results Compact bone derived-MSCs were fibroblast-like cells,positive for Sca-1,CD44,CD29,and CD105,but expressed no CD34,CD45,CD11b and CD31.They could differentiate into adipocytes and osteoblasts in vitro.Compact bone derived-MSCs infused by tail vein were capable of protecting lung from injury.In BLM+MSCs group,2.5?105 MSCs infusion demonstrated more efficient protective activity than that of 5?105 MSCs;the lung injury was less severe in BLM+BUS+MSCs group than that in BLM +MSCs group.The content of hydroxyproline in lung tissue of normal,BUS,BLM,BLM+BUS,BLM+MSCs 1,BLM+MSCs 2,BLM+BUS+MSCs 1 and BLM+BUS+MSCs 2 group was 0.72?0.05,0.79?0.10,1.37?0.09,1.22?0.16,1.24?0.14,1.01?0.15,1.03?0.19 and 0.98?0.12?g/mg,respectively.The content of hydroxyproline increased in BLM and BLM+BUS group compared with that in normal group(P

ABSTRACT

Objective To observe the expression of anti-cardiac myosin antibody(AMA) and the collagen volume fraction(CVF) in serum in mice with chronic viral myocarditis(CVMC),and to explore the preventive and protective function of oxymatrine on myocardium.Methods BALB/c mice(n=60) were infected with coxsackievirus B3(CVB3) of increased dose biweekly to establish CVMC model.Mice in normal control group(n=8) received equal-volume 9 g?L-1 saline without CVB3 at the same time.The surviving mice in CVMC model group were randomly divided into CVMC control group(n=8) and oxymatrine therapy group(n=8) at the 42th day.From then on,mice in oxymatrine therapy group were treated with oxymatrine at the dose of 100 mg?kg-1?d-1 by gastric perfusion once a day for 28 days,and meanwhile mice in CVMC control group and the normal control group received equal-volume 9 g?L-1 saline by gastric perfusion every day.Then all mice were sacrificed at the end of the experiment.The ratio between heart weight to body weight(HW/BW) was calculated.Myocardium slides were stained with collagen specific Picric acid-Sirius red staining,and the CVF was calculated with image analysis software.The serum level of AMA(optical density value,A value) was detected by enzyme-linked immunosorbent assay(ELISA).Results HW/BW(0.007 9?0.000 3),CVF [(15.30?1.08)%] and the A value of AMA(0.286?0.053) in mice of CVMC control group were increased significantly compared with those in normal control group HW/BW(0.005 5?0.000 2),CVF[(6.84?1.11)%],the A value of AMA(0.160?0.050)(Pa
